Mitochondrial Sequence Analysis based on Mass Spectrometry

Mass spectrometry sequencing refers to the determination of the amino acid sequence of the sample's primary structure using mass spectrometry. The sequence analysis of proteins, antibodies, peptides, and other samples based on mass spectrometry analyzes the amino acid sequence of mitochondrial samples.

Compared with Edman's degradation, mass spectrometry sequencing is more sensitive. It can cleave peptides more quickly, and can recognize terminal blocked or modified protein samples. Using the existing high-resolution mass spectrometry technology platform, Creative Biogene provides sequence analysis services based on mass spectrometry, which can cover 100% of the determined mitochondrial target protein sequences.

In the process of protein sample sequence analysis, only Trypsin is used to digest the protein, and the coverage rate of the identified peptide is about 60%. To obtain all the sequence information of the target protein, Creative Biogene selected six kinds of proteases (Trypsin, Chymotrypsin, Asp-N, Glu-C, Lys-C, and Lys-N) commonly used in the process of protein sequence analysis to determine the target protein by restriction endonuclease digestion and mass spectrometry. At the same time, 100% of the protein sequence can be determined by splicing between the peptides.

Creative Biogene uses Thermo's Orbitrap Fusion Lumos mass spectrometer to analyze the sequence of protein samples. Orbitrap Fusion Lumos mass spectrometer is now the highest resolution and sensitivity mass spectrometer, which ensures the sensitivity of the identification of low-abundance peptide fragments. At the same time, it adopts the binding mode of HCD and ETD in the process of peptide fragmentation to ensure the integrity of peptide fragments. It can realize N-terminal and C-terminal sequence analysis and full-length protein sequence analysis of protein samples. The samples with the unknown theoretical sequences can be analyzed by ab initio sequencing.
