Succinate dehydrogenase (SDH) is widely found in animals, plants, microorganisms and cultured cells.SDH is a marker enzyme of mitochondria, a membrane-bound enzyme located on the inner mitochondrial membrane, and is one of the hubs linking respiratory electron transfer and oxidative phosphorylation, providing electrons for the productive respiratory chain of many prokaryotic cells.SDH catalyzes the dehydrogenation of succinic acid to produce fumaric acid and the reduction of 2,6-dichlorophenol indophenol (DCPIP) through the transfer of phenazine dimethyl sulfate (PMS), and has a characteristic absorption peak at 600 nm. The rate of reduction of 2.6-DPIP was measured by the change of absorbance at 600 nm, which represents the SDH enzyme activity. |
