Protein unlabeled quantification (label-free) is a mass spectrometric analysis of protein hydrolyzed peptides by LC-MS/MS technology. It does not use expensive stable isotope tags as internal standards. It works by analyzing the mass spectrometry data produced during large-scale protein identification and comparing the corresponding peptides' signal intensity in different samples to quantify the proteins corresponding to the peptides. Compared with iTRAQ technology, the sample volume required for label-free technology is small, and the cost is low. The label-free technique can be used for quantitative comparative analysis of high-throughput biomarker screening, protein-protein interaction, protein post-translational modification, and so on.
